About Foundayo®

Foundayo® (orforglipron) is an oral, once-daily GLP-1 receptor agonist approved by the FDA in April 2026 — the first GLP-1 pill cleared for chronic weight management in adults. As a small-molecule tablet rather than a peptide, it can be taken any time of day with or without food or water restrictions, and it does not require refrigeration. What the studies show

Foundayo® was studied in Eli Lilly's ATTAIN and ACHIEVE phase 3 programs. The ATTAIN program evaluated orforglipron for chronic weight management; at the highest doses, completers averaged roughly 12% body weight reduction along with improvements in waist circumference, cholesterol, triglycerides, and systolic blood pressure. ACHIEVE-3 compared orforglipron to oral semaglutide in adults with type 2 diabetes inadequately controlled by metformin, with orforglipron producing greater weight loss and glycemic control.

Foundayo® binds to and activates the GLP-1 receptor in the brain and gut. This reduces appetite, slows how quickly your stomach empties, and supports glucose regulation. The result is smaller portions feeling more satisfying, longer-lasting fullness between meals, and gradual weight loss.

  • Known hypersensitivity to orforglipron or its components
  • Patients relying on oral contraceptives without backup birth control — Foundayo® can reduce the effectiveness of oral birth control for ~30 days after starting and after each dose increase

Safety & side effects

Foundayo® shares the gastrointestinal side-effect profile of the GLP-1 class. Most effects are most noticeable around dose increases and ease with time.

Common

  • Nausea, especially during titration
  • Diarrhea or constipation
  • Bloating or reflux
  • Decreased appetite
  • Mild headache

Less common but serious

  • Pancreatitis — severe, persistent abdominal pain
  • Gallbladder disease, including gallstones
  • Kidney injury from dehydration related to GI side effects
  • Possible thyroid C-cell tumors (class warning based on rodent studies)

Dosing

How to take Foundayo

  • Foundayo® is taken as one tablet by mouth once daily. The standard titration starts at 0.8 mg, then steps up to 2.5 mg, 5.5 mg, 9 mg, 14.5 mg, and a maintenance dose of up to 17.2 mg — with at least 30 days at each level based on response and tolerability. Your provider chooses the maintenance dose that works best for you. Specific dose is set by your provider based on goals and tolerance.

    How to take

    1. 1Take at roughly the same time each day; food and water timing are flexible.
    2. 2Swallow the tablet whole — do not split, crush, or chew.
    3. 3If you use oral birth control, your provider may recommend a backup method for ~30 days after starting Foundayo® and after each dose increase.
    4. 4If you miss a dose, follow the instructions on your prescription label or contact your provider. Do not double up.
